Tesamorelin is a synthetic 44-amino-acid analogue of growth hormone-releasing hormone carrying a trans-3-hexenoyl group at its N-terminus, a modification that confers resistance to dipeptidyl peptidase IV cleavage.

TesamorelinMetabolic Pathway Peptides

Overview

Tesamorelin is a synthetic analogue of growth hormone-releasing hormone that reproduces the full 44-amino-acid sequence of the parent hormone, GHRH 1-44. A trans-3-hexenoyl fatty acid group is attached at the N-terminal tyrosine, giving the molecule resistance to cleavage by dipeptidyl peptidase IV. Its full-length sequence distinguishes it from the shorter GHRH fragments sermorelin and CJC-1295.

In vitro, tesamorelin acts as an agonist at the GHRH receptor expressed on pituitary somatotrope cells, a class B G-protein-coupled receptor that signals through adenylate cyclase and cyclic AMP. The compound is supplied as a lyophilized powder in a 20 mg vial.

Mechanism of Action

Tesamorelin is an agonist at the GHRH receptor, a class B G-protein-coupled receptor on pituitary somatotropes; receptor occupancy activates adenylate cyclase and raises intracellular cyclic AMP.
