The pentapeptide portion of Palmitoyl Pentapeptide-4 corresponds to a fragment of the C-terminal propeptide of type I procollagen, placing it in the matrikine class of extracellular matrix-derived peptide fragments. N-terminal conjugation to palmitic acid gives the molecule a lipophilic tail.
In cultured fibroblasts the compound has been reported to upregulate transcription of type I and type III collagen and of fibronectin and to alter glycosaminoglycan output. The lipopeptide is sparingly soluble in water and dissolves in polar organic solvents such as dimethyl sulfoxide.
Mechanism of Action
In cultured fibroblasts the peptide acts as a matrikine signal, upregulating expression of type I and type III collagen and of fibronectin in vitro; the palmitoyl chain confers lipophilicity to the molecule.
