SNAP-8, also designated Acetyl Glutamyl Heptapeptide-1, is a synthetic octapeptide whose sequence corresponds to the N-terminal region of SNAP-25, a component of the SNARE protein complex. It extends the hexapeptide analogue Acetyl Hexapeptide-3 (Argireline) by two residues; terminal acetylation and amidation neutralize the free charges at both ends of the chain.
In vitro, the peptide competes with SNAP-25 for the SNARE assembly interface, occupying that binding surface without proteolytic cleavage of the parent protein. The compound is hydrophilic and dissolves readily in aqueous buffer.
Mechanism of Action
Competitively occupies the SNAP-25 N-terminal binding interface of the SNARE complex in vitro, reducing assembly of the complex without cleaving SNAP-25; the interaction is non-proteolytic and reversible.
