Overview

Glutathione (gamma-L-glutamyl-L-cysteinyl-glycine) is a tripeptide assembled intracellularly from glutamate, cysteine, and glycine by gamma-glutamylcysteine synthetase and glutathione synthetase. The gamma-glutamyl linkage at the N-terminal residue is not recognized by most peptidases.

The molecule exists in reduced (GSH) and oxidized (GSSG) forms that interconvert through thiol-disulfide exchange, and the GSH:GSSG ratio is a measured index of redox state in cultured cells. GSH is the substrate of glutathione S-transferases, which catalyze conjugation to electrophilic compounds. The lyophilized solid is freely soluble in water.

Mechanism of Action

The reduced thiol donates electrons in glutathione peroxidase-catalyzed reactions, and the same thiol serves as the conjugating group in glutathione S-transferase-catalyzed phase II conjugation reactions in vitro.
