IGF-1 LR3 (Long R3 IGF-1) is an engineered analogue of insulin-like growth factor 1. Two modifications distinguish it from the native 70-residue sequence: glutamate at position 3 is replaced by arginine, and a 13-residue extension is appended to the N-terminus, giving a total length of 83 amino acids.
The position-3 substitution lowers affinity for the insulin-like growth factor binding proteins (IGFBP-1 through IGFBP-6), and the N-terminal extension slows proteolytic cleavage. The analogue remains an agonist at the IGF-1 receptor (IGF1R) and is supplied as a lyophilized powder.
Mechanism of Action
Agonist at the IGF-1 receptor (IGF1R); receptor engagement activates PI3K/Akt and MAPK signaling in cultured cells. The position-3 arginine lowers IGFBP affinity relative to native IGF-1.
