Overview

Melanotan-II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone with the structure Ac-Nle-cyclo(Asp-His-D-Phe-Arg-Trp-Lys)-NH2. An amide bond between the aspartate and lysine side chains forms a lactam bridge that constrains the ring conformation and gives the molecule resistance to enzymatic degradation. It is the structural parent of bremelanotide.

In vitro the peptide is a non-selective agonist at melanocortin receptors MC1R, MC3R, MC4R, and MC5R, each a Gs-coupled receptor coupling to adenylate cyclase and cyclic AMP. Melanotan-II is supplied as a lyophilized powder in an 8 mg vial.

Mechanism of Action

Melanotan-II is a non-selective agonist at melanocortin receptors MC1R through MC5R; its lactam-constrained ring conformation is associated with high measured binding affinity at each Gs-coupled subtype in vitro.
