Overview

PNC-27 is assembled from two segments. The N-terminal segment reproduces residues 12-26 of the p53 transactivation domain, the region that forms the contact surface with HDM-2. The C-terminal segment is a cationic, tryptophan- and arginine-rich sequence of the penetratin class, which gives the construct affinity for lipid bilayers.

The construct carries a net positive charge at neutral pH. Published molecular characterization originates from a small and closely related set of research groups; no independent replication was identified, and the compound holds no approved status in any jurisdiction. It is supplied strictly as a laboratory reagent.

Mechanism of Action

The p53-derived segment corresponds to the region of p53 that contacts HDM-2; the cationic penetratin-class segment associates with lipid bilayers in vitro. No further mechanism is claimed for this reagent.
