SS-31, also designated elamipretide and MTP-131, is a synthetic tetrapeptide with the sequence D-Arg-dimethylTyr-Lys-Phe-NH2. The alternating aromatic-cationic residue pattern gives the molecule a net positive charge and membrane permeability, and the peptide partitions into the inner mitochondrial membrane, where it associates with the phospholipid cardiolipin.
Cardiolipin association is a physicochemical interaction with the lipid headgroup rather than an enzyme-inhibition event, and in vitro the peptide accumulates in the mitochondrial compartment relative to surrounding buffer. Supplied as 10mg of lyophilized powder per vial.
Mechanism of Action
Associates with cardiolipin in the inner mitochondrial membrane through charge and hydrophobic interactions, a binding event linked in vitro to the assembly state of electron transport chain complexes.
